摘 要 经一系列试验证实百日咳菌大罐培养浓度的下降与培养基中不耐热营养因子不足有关,将培养基的高压灭菌方式改为除菌过滤,保留热不稳定营养因子,可以提高百日咳菌大罐培养浓度;优化酸沉淀条件后,提高了菌苗回收率,从而节省了大量培养基,缩短了生产时间,获得了良好的经济效益。关键词 百日咳菌苗 大罐培养 高压灭菌 除菌过滤分类号 R378.42Application of Media Filter Sterilization to the Production of Pertussis Vaccine L iu Baokui,L iu W enda,Zhou Di,et al.( N ational V accine &Serum I nstitute,Beij ing 100024)Abstract T he decrease of fermenter culture concentration of pertussis bacteria is related to the insufficiency of heat-labile grow th factor in media confirmed by a series of experiments.T he bacteria concentration in fermenter culture w as increased by means of sterilizing the media w ith filtration w hich retained the heat-labile grow th factor instead of autoclave,and the rate of culture recovery w as improvedby the optimization of acid precipitation conditions.T herefore,a good economic result is gained by savingboth the production time and the amount of media used in the production of pertussis vaccine.Key words Pertussis vaccine Fermenter culture Autoclave Sterilization Filter sterlilzation
